9/14/10 Pharm 1001 Sleep study – most people would be within the middle 68% Some sill be really sensitive and some will be resistant to the drug Why are there such big differences? - absorption distribution biotransformation excretion --- everyone does it differently - Age o “normal”/average = 12(18) – 65 (in reasonable health o 60-90kg (130-200lbs) Infants - dosage due to how tall the person is and how much they weigh = BODY SURFACE AREA - infants are no small adults - inability to pharmacokinetically handle drugs o poorly developed drug metabolic enzyme systems o kidneys: functionally inefficient pharmacodynamic differences – Elderly individuals - >65 group = 12% population o 25% Rx - >60 = 17% population o 39% hospitalizations o 51% all drug related fatalities ability to eliminate drugs are decreased - Drug elimination: kidney and liver o Excretion processes reduced o Drugs remain in body longer o Pharmacodynamic changes drug response? Body Weight
