BIO 119 McCague File-014 Exercise III Purpose: To determine the red and white cell count and blood types in normal human adult. Materilals: 1. Hemocytometers (Precision instruments. Handle with care) 2. Red cell dilution pipettes. 3. White cell dilution pipettes. 4. 3% NaCl, or Hayem’s Solution. 5. Special solution for white cells. 6. Wright’s stain. 7. Clean slides. 8. Hemolets 9. Distilled H 2 O. 10. Anti-Sera for typing. Procedure: A. Determination of Red Blood Cell Count 1. Blood secured after puncture by a hemolet from a dried finger previously cleaned with alcohol is drawn up carefully to the 0.5 mark on the larger of the two pipettes or the red cell dilution pipette. If it is drawn up above the mark a little may be blown out. Then the cleaned end of the pipette is quickly placed under the diluting fluid and drawn up to 1.01 which means diluting the blood 1:200. Next the pipette is held between the thumb and a finger and the contents shaken until properly mixed. The first few drops are blown out. A drop is placed on the center of the
