File-070 McCague Introduction to Muscle Cell Contraction Physiology The materials you will need to perform this lab exercise include: Rabbit Psoas Muscle “Living” preserved in cold glycerine; ATP solution in dropper vial, salts in dropper vials, millimeter ruler, clean sides and coverslips, teasing probe, microscope. Procedure: 1. Remove psoas muscle from the tube and place it in a dish. Pour the glycerine in the tube over the muscle. 2. Tease out a single muscle fiber “cell” at least 10mm long and mount it on a microscope slide. Add a cover slip. 3. Measure the length of the muscle fiber in millimeters. Initial length- _______mm. 4. Look at the transparent muscle under the microscope. Note the striations and the nuclei. 5. Flood the slide near the edge of the coverslip with solution marked ATP + Salt. Do this while your partner is looking through the microscope. Did the striations
