Unformatted text preview: Linkage Mapping Feb 14 th , 2011 Announcements 3 rd problem set due Friday, 4:00 pm Review RECOMBINATION can occur as a result of independent assortment or crossing-over. RECOMBINANT gametes have a different genotype than either haploid genomes of the parent. Genes on the same DNA molecule are LINKED. CROSSOVERS can disrupt linkage. RECOMBINATION FREQUENCY varies with the distance between linked genes. Recombination INPUT MEIOSIS OUTPUT Recombination has occurred if the genotype of a gamete is different than the input haploid genotypes. o INDEPENDENT ASSORTMENT o CROSSING OVER Mapping with two-point testcrosses crosses RF A and B 16 % A and C 5 % A and D 50 % B and C 21 % B and D 50 % C and D 50 % hat is the order and distance between four markers A, B, C & Mapping with two-point testcrosses crosses RF A and B 16 % A and C 5 % A and D 50 % B and C 21 % B and D 50 % C and D 50 % hat is the order and distance between four markers A, B, C & A B 16 cM Mapping with two-point testcrosses crosses RF A and B 16 % A and C 5 % A and D 50 % B and C 21 % B and D 50 % C and D 50 % hat is the order and distance between four markers A, B, C & A B 16 cM C 5 cM Mapping with two-point testcrosses crosses RF A and B 16 % A and C 5 % A and D 50 % B and C 21 % B and D 50 % C and D 50 % hat is the order and distance between four markers A, B, C & A B 16 cM C 5 cM remember independent assortment will result in a 50% recombination frequency (unlinked) Mapping with two-point testcrosses crosses RF A and B 16 % A and C 5 % A and D 50 % B and C 21 % B and D 50 % C and D 50 % hat is the order and distance between four markers A, B, C & A B 16 cM C 5 cM D Testing Linkage AaBb X aabb 93 Aabb 108 AaBb 104 aabb 95 aaBb note: nomenclature does not indicate if genes are linked from these test cross ratios can you tell if genes are unlinked?...
