Subitilis is gram positive; it is going to be hard to separate the DNA - Proteinase K: is a detergent - SDS: - Ribonuclease A: breaks down in the RNA - Lysozyme: breaks down in the enzyme wall - Clean three times We isolated everything; the entire genome of the subitilis is there and that why need the PCR which in every cycle How does the PCR work? - Polymerase chain reaction/endpoint PCR; Helix case need to separate the two DNA strands; we will need to add 95 % heat which will break it off A heat stable will be used because of the substance lack to handle 95% heat; enzyme that is used to synthesize a small DNA sequence of interest within a complex starting DNA (e.g. total genomic DNA)
