Lab 6 Control of organogenesis in cultures of Nicotiana tabacum Objective: To observe the effects of various levels of hormones on tobacco explants. Protocol: 1. You will be given a container with several tobacco seedlings. The seedlings have been grown from sterilized seed grown aseptically, so the tissue will not have to be disinfested. 2. Using sterile technique, and a scalpel and forceps, remove the cotyledons and carefully place 6 onto each plate provided. There are Four media types (listed below) and you have two plates of each type. 3. Seal the plates with Parafilm. Label each plate.

Unformatted text preview: 4. Examine your explants at weekly intervals and record your observations. One set of plate will be kept under light and the other set will be kept in the dark. Be sure to label each plate either light or dark. Media: MS media + additives BA(mg/L) NAA(mg/L) 2.0 0.2 0.2 2.0 1.0 1.0 *the plates have been marked with a number of lines for 1, 2, 3 & 4. This number does not correlate to the media listed above. It is up to you to determine which media type with which based on how the tissue behaves over time....
