Abstract : Absorbance readings (420 nm) were taken for different enzyme concentrations. The higher the absorbance meant the more concentration present. A spectrophotometer was used to calculate the absorbance readings. The first portion of the experiment tested to find the relationship between enzyme concentration and enzyme activity. The next part of the experiment tested the effects of substrate concentration on enzyme activity. The relationship between temperature and enzyme activity was then tested. Lastly, different pH levels were tested by using absorbance readings to determine their effects on enzyme activity.

Main Introduction: Enzymes are proteins known as catalysts that lower the activation energy required for a chemical reaction; therefore, the chemical reaction occurs at a faster rate. These catalysts have a lock-and-key fit (Campbell 2008). This means that only a particular substrate can bind at the active site with a particular enzyme. When enzymes lose their shape or unfold, they are said to be denatured and no longer work properly (Harris-Hallar 2010). Changes in substrate concentration, enzyme concentration, temperature and pH all affect enzyme activity (Harris-Hallar 2010). A spectrophotometer is used to measure absorbance levels. The higher concentration of a particular molecule in a solution, then there will display higher absorbance (Harris-Hallar 2010). The spectrophotometer can only compare absorbance and transmittance of different substances within cuvettes; therefore, a blank must always be used before hand to get a one hundred percent transmittance reading first to in return get an accurate absorbance measure for the substances being tested (Harris-Hallar 2010). To record absorbance of a substance on the spectrophotometer, a person must zero it out on transmittance, place in a blank, bring it to 100, replace the blank with the substance being tested and then hit the absorbance button.
