Josh Laster 523 – Biology 111 10/25/10 Amount of Absorbance for the Four Main Photosynthetic Pigments Abstract: Use a spectrophotometer to take absorbance readings of carotene, chlorophyll a, xanthophyll, and chlorophyll b from 400 nm to 700 nm in 50 nm increments. Use a blank for proper comparison. Set the filter lever for wavelengths of 340 to 599 nm (black scale). The results show that a wavelength of 450 nm has the best absorbance for every type of pigment tested. This concept is illustrated on the action and absorption spectra because the most oxygen production is produced at 450 nm. The Carotene pigment mostly absorbed wavelengths from violet to green (400 nm – 550 nm). Chlorophyll a pigment absorbed mostly the violet through blue wavelengths (400 nm – 480 nm) and yellow through red (600 nm – 680 nm). As absorbance increases, oxygen production is greater because there is more energy for photosynthesis to take place (relationship between the action and absorption spectra). There is more energy available in the shorter wavelengths; therefore, the oxygen output at 450 nm is the greatest on the action spectra.
