DNA Mutation and Repair: Mutation is defined as an inheritable change in a chromosome. After replication the altered base is permanently incorporated into the genome. Two types of mutations: Base: Point mutations- substitute one base for another Missense- change an amino acid codon and protein synthesis Nonsense- change an amino acid codon to a stop codon Frameshift: change of translational reading frame Base excision- removal of single nucleotide Nucleotide excision: removal of group of nucleotides Carcinogens like ethidium bromide can cause mutations including base insertions Principles of Thymine-dimer Repair: First endonuclease recognizes the distortion cause by the thymine dimmer. The DNA around the thymine dimmer is displaced by DNA polymerase I. DNA ligase seals the ends Mismatch repairs correct mistakes made my DNA Polymerase RNA structure: Ribosomal RNA (rRNA) : is the most abundant Transfer RNA (tRNA) : one or more exists for each amino acid Messenger RNA (mRNA) : although the least abundant, is specific for each protein
