V(D)J Recombination Purpose: V(D)J recombination generates diversity for an adaptive immune response. V(D)J recombination: RAG1 and RAG2 direct chromosomal breakage at receptor gene loci. Nonhomologous end joining (NHEJ) resolves chromosome breaks to assemble a mature receptor gene (immunoglobulin or T cell receptor) and deleted extrachromosomal circle. Yellow rectangles denote receptor coding DNA, and triangles denote recombination signal sequences (RSS).

Unformatted text preview: Important Note: TdT (terminal deoxynucleotidyl transferase) adds nucleotides randomly to the ends of the segments to be ligated. This provides junctional diversity. After the random nucleotides have been added the ends are joined by NHEJ (non-homologous end joining). Figure adapted from Reddy et al. V(D)J Recombination: Born to be Wild. 2010.
