Lab 3 Worksheet: Stormy Williams; Krause-208 September 22, 2011 1. To calibrate the eyepiece micrometer a clean and clear stage micrometer was necessary. The stage micrometer was placed on the stage of the microscope and focused so that the lines on the stage micrometer were clearly seen. Then the first line of the stage micrometer was aligned with the first line of the eyepiece micrometer. Then, the observer determined where the lines were again aligned. To determine the calibration factor for 100x; the number of stage spacings (10) was multiplied by the distance per stage space (a constant of 10). Then, the product of those two numbers (100) was divided by the number of eyepiece spacings (10). The “unit” on eyepiece was 10 micrometers per eyepiece space. The microscope objective was then rotated to the 40x objective lens and focused so that the lines on the stage micrometer were clearly seen by the observer.
