Exercise 1-2 Tryptic Soy Broth (TSB) and Tryptic Soy Agar (TSA) Preparation NOTE: Tryptic Soy Agar (TSA) and Broth (TSB) will replace the Nutrient Broth and Agar referred to in the lab manual. TSA and TSB are rich undefined media that can be purchased as a dehydrated powder. EACH PAIR OF STUDENTS WILL MAKE TSA PLATES NOTE: Due to time constraints your flask of agar to make your TSA plates has already been prepared and autoclaved. You will begin at STEP #5 of the procedure. To prepare TSA in flasks for pouring into Petri dishes use the following recipe: Ingredient Quantity Dehydrated Tryptic Soy Agar 6 grams DI-H 2 O (DI = deionized) 150 ml 1) Using the triple-beam balance, weighing paper and spatula, weigh out the media powder and transfer it into a 250 ml Erlenmeyer flask. 2) Using a graduated cylinder measure out 150 ml of DI-H 2 O and add the water to the flask. Gently swirl the media to disperse the powder. Because the media contains agar it will not dissolve before it is autoclaved. Cover the top of the flask
