Lab Notebook Example Page 1 Ex 1-2 Tryptic Soy Broth (TSB) and Tryptic Soy Agar (TSA) Preparation August 23, 2010 Purpose: To learn how to prepare microbiological media Methods: The procedure outlined in Ex 1-2 “Nutrient Broth and Nutrient Agar Preparation” p 14 Microbiology Laboratory Theory & Application 3 rd edition, Leboffe and Pierce was performed with the following modifications. Tryptic soy broth and agar were used in place of nutrient broth and agar. Tryptic soy broth and agar are undefined media composed of tryptone, soytone, sodium chloride and dipotassium phosphate (Microbiology Laboratory Theory & Application 3 rd edition, Appendix H, p 470). The TSA and TSB media was prepared as per the Media Recipe Supplemental Material posted on D2L. Briefly, for the TSA media 6 grams of tryptic soy agar was added to 150 ml of water in a 250 ml flask and then the media sterilized for 20 minutes at 121 ° C and 15 pounds pressure in the autoclave. Following autoclaving the media was cooled to ~50°C and then used to pour plates.
