11 Sub-culturing Bacterial Isolates Purpose: To obtain a PURE culture of a bacterial species Why is this so important? Without a pure culture NO morphological, physiological or biochemical analysis can be done since contaminating bacterial species will confound all test results. 22 Obtaining a Pure Culture How many different colony types? Each colony type may indicate a different bacterial species. To identify any one bacterial species a single colony must be “ picked ” and separated from the rest of the bacterial species on that plate. MBIO 3812

33 Obtaining a Pure Culture Each colony type needs to be WELL ISOLATED to ensure it’s the ONLY colony touched by the inoculating loop. The bacteria in that colony is then quadrant streaked for isolation.
