Unformatted text preview: Genetics 380, Spring 2008 Examination #3 Answers Q. 1. (10 pts) A student sets up a DNA sequencing reaction, but pipets the incorrect reagents into different tubes. All the reactions contain DNA polymerase, template and Primer. However, each reaction contains the indicated nucleotide mixes (dNTP indicates deoxynucleotides, ddNTP indicates dideoxy-nucleotides). The student performs the reactions shown, runs the DNA products out on a sequencing gel, and analyzes the gel. Please draw the expected DNA bands on the sequencing gel below. The first two lanes have been done for you as an example. Q. 2. (12 pts) A circular DNA molecule carries the Ampicillin-resistance (AmpR ) gene. You perform single and double digests with the indicated restriction enzymes, and you run the DNA out in two agarose gels. You stain one of the gels with ethidium bromide so that you can see all of the DNA bands (panel on the left). You transfer the DNA out of the other gel onto a filter and perform a Southern blot using sequences for the AmpR gene as your hybridization probe (the resulting autoradiogram is the panel on the right). The sizes, in kilobases, are indicated for each band. a. (4 pts ) For each restriction enzyme, indicate how many cutting sites are there on the circular molecule.
