March 30, 2011 The Determination of the Retention Index in Organic Compounds using n-Alkanes Background In chromatography, the separation of the components of a mixture is the result of the different interactions that exist between the (i) solute and the mobile phase, (ii) the solute and the stationary phase, and (iii) the stationary phase and the mobile phase. The ability of the method to separate the mixture into zones of each solute is determined by the different strength of these interactions of the several components of the mixture for a given stationary phase and mobile phase combination. In GC (gas chromatography), the mobile phase is an inert gas and the ability to separate the components of a mixture is a result of the solute stationary phase interactions. To identify the importance of different intermolecular interactions between the separation of solutes and the stationary phase we can use the Kovat’s Retention Index and the McRynold’s Numbers. Kovat’s showed there is a linear relationship between the defined Retention Index and the Logarithm of the adjusted retention time for n-alkanes. N-alkanes can only interact with the stationary phase by van der Waals forces. The additional carbon atoms in an n-alkane series allows stronger van der Waals interactions between solute and the stationary phase and, therefore, longer retention
