Amir Jabbari Period 2 10/16/08 Enzyme Lab Question: How does pH affect the activity of the enzyme catalase? Background Info: Catalase breaks down hydrogen peroxide (poison that builds up in cells) H2O2 (Hydrogen Peroxide) H2O (Water) + O2 (Gas) Balanced Form: 2 H2O2 2 H2O + O2 Hypothesis: If catalase activity is affected by pH then I predict it will be greatest when it is very acidic. Materials: 10 Test tubes Catalase Enzyme Acid Ruler Hydrogen Peroxide Base Stirring rod Masking tape pH paper Methods: 1. Use 1 ml. of enzyme solution at it’s normal pH which is 5-6 2. Raise 1 ml. of enzyme solution to a pH of 9-10 by adding 1 drop of NaOH 3. Raise 1 ml. of enzyme solution to a pH of 11-12 by adding 2 drops of NaOH 4. Lower 1 ml. of enzyme solution to a pH of 11-12 by adding 2 drops of NaOH 5. Lower 1 ml. of enzyme solution to a pH of 1-2 by adding 2 drops of HCl. 6.
