Lab 9 Page 1 Lab 9 Lab Clean-Up (Page 2) Test Tubes, Test Tubes and more Test Tubes. Just please be sure and remove the Tape from all of these Test Tubes before you put their Caps in the Wire Basket and the Tubes themselves in the Metal Bus Pan. You’ll probably have Plates for Oscar. Some of you may have Slides, Toothpicks and Filter Paper. Be sure everything gets put in the appropriate Place. Lab Exercises 1- Biochemical Tests Results (Pages 3-5) It’s Test Tube City. Some of the Biochemical Tests are really easy to score (Phenol Red Mannitol, Citrate) but some take a bit of “developing” (particularly the Voges-Proskauer, which takes ~45 Minutes). Good Organization and good Bookkeeping are really important here. And compare your Results with other Students’ Results. 2- Biochemical Unknowns Results (Page 6) There are two Approaches here. On the one Hand, you could “run” your Clan’s Unknown Biochemical Test Results in parallel with your Isolations Biochemical Test Results and do Real Time Comparisons. And if you don’t have an IMViC this is probably the best Choice. On the other Hand, you can “run” your Unknown Biochemical Test Results after you’ve practiced using your Isolations Biochemical Test Results. This will take a bit longer 3- Background Information for Biochemical Testing (Pages 7-12) 4- Blood Smears and Wright ʼ s Stain (Pages 13-17) You’ll be performing a Wright’s Stain on Blood Samples. You’ll should be able to see Red Blood Cells and you’ll be able to distinguish some Inflammatory Cells and Lymphocytes.

Lab 9 Page 2 Lab 9 Clean-Up Once again we’ve got a whole Bunch of Stuff that needs to be disposed of properly. You’ve probably been doing this as you work, but just in case, here’s the Deal: Biochemical Testing and Biochemical Unknowns • Test Tubes go into the Aluminum Bus Pan - Please remove all Tape - Caps go into the Wire Basket in the Aluminum Bus Pan • Plates go into Oscar - It’s OK to leave the Tape on your Stacks of Plates • Slides (Gram Stain and Wright’s Stain) go into the Metal Pitcher • Used Toothpicks go into this same Waste Beaker • Unused Toothpicks (still in the Beaker) go back into the Basket from which you took them. - We can autoclave these guys and re-use them. • Filter Paper goes to Oscar - It’s contaminated with Bacteria Lab Benches and Drawers • Please check your Lab Drawer for any Plates, Tubes, Flasks (which shouldn’t have been in there in the First Place!) or Personal Articles • Please remove the Tape with your Name from your Lab Bench Edge !
