Lab Exercises Week # 5 Inoculation of exercise 2-6 Fluid thioglycolate media This media is designed to permit the growth of organisms with their optimal level of oxygen whether that be need ( strict aerobe ), tolerance ( aerotolerant ), ambivalence ( facultative anaerobe ) or strict absence ( strict anaerobes ) Materials: 4 test tubes containing FT media Broth cultures of: Pseudomonas aeruginosa Clostridium sporogenes Staphylococcus aureus When inoculating, dip the loop to the bottom of the FT media and GENTLY mix the media with the loop as you are removing it. Purpose ? Test tube #4 is your negative control. Incubation temperature 37 o C Time: 48 hours Exercise 2-7 Anaerobic Jar Inoculate 2 agar plates using the same organisms from the previous lab activity. One plate will be grown in the 37 o C incubator in the anaerobic jar and the other plate will be placed as we normally do into the green tray to the 37 o C incubator. Anaerobic jar chemical reactions when water is added: NaBH 4 + NaHCO 3 + H 2 O H 2 + CO 2 H 2 + O 2 * + Pd (catalyst) 2 H 2 O
