1 Joseph Vallillo Section: F1 10/19/2011 THE EFFECTS ON ALKALINE PHOSPHATASE DUE TO TEMERATURE AND pH ABSTRACT: The effects on the rate of reaction of alkaline phosphatase on the substrate p-Nitro-Phenyl Phosphate (pNPP) due to temperature and pH were tested. Enzyme-substrate reaction was assayed at temperatures ranging from 0 ° C to 100 ° C. It was assayed again at pH levels ranging from 2 to 12. An optimal rate of reaction occurred at boiling temperature of water, 100 ° C for the first assay. For the second assay the rate of reaction was optimized at an alkaline pH level of 10. INTRODUCTION: In this experiment we looked at the rate of reaction of the enzyme alkaline phosphatase on the substrate p-Nitro-Phenyl Phosphate. A catalyst is a “protein catalyst that affects the speed of the reaction without being consumed by the reaction,” whereas the substrate is the “substance on which [the catalyst] works” (Solomon et al ., 2011). The resulting substance is the product. In this experiment the product is p-Nitrophenol (pNP). Catalysts work by lowering the activation energy of the reaction, “the energy required to break the existing bonds and begin the existing reaction” (Solomon et al ., 2011).
