Richard Duong MCRO Lab 9 September 2010 Week 2 Pre-Lab Purpose: to learn and practice the aseptic techniques by making slides of different microorganisms in the first part of the lab. In the second part of the lab, a gram stain will be performed on a slide of different microorganisms. Procedure: Aseptic Technique 1. Turn on incinerator and allow it to warm up for several minutes 2. Each student takes two glass slides, marks each with a nickel-sized circle with a wax pencil, and applies a piece of tape to both sides (covering ¼ inch of the slide and ¾ inch of tape serving as a handle) 3. Place a small loopful of water to the slide 4. Inoculating loop should be heated to bright red by holding it down the incinerator (reddish-orang on the inside) a. The incinerator will burn quickly, so watch your hand 5. After loop is red hot, allow 10-15 seconds to cool 6. Lightly touch the colony (we don’t want too many or else the smear will be hard to interpret) and transfer it to a slide. 7.
