Richard Duong MCRO Lab 14 September 2010 Week 3 Pre-Lab Purpose: To learn and practice the techniques of broth and agar culture transfers. Within this lesson, we will also learn the proper streak plating technique for bacterial isolation. In addition, a gram stain will be prepared from a broth, differing from last week’s dry preparation of a gram stain. Finally, we will view demonstrations based on oxygen requirements of Eschrichia coli , Micrococcus luteus , and Clostridium perfringens while also viewing anaerobic culture. Procedure: A. Aseptic technique with culture tubes 1. Every student needs to make two transfers—one to each media (broth and agar slant). 2. Tie up any loose hair and sleeves, make sure to avoid any unnecessary movement. 3. Loosen any caps that may be tightly screwed. 4. Heat the inoculating loop to be bright red, allow to cool for 10-15 seconds. 5. Holding a tube with your left hand, remove cap with 4 th and 5 th fingers of right hand; NEVER LAY CAP DOWN—keep at semi-horizontal angle to prevent dust from
