Richard Duong MCRO 251L 13 September 2010 Week 7 Pre-Lab Purpose: To complete exercises from last week and fulfill new objectives.There are two new objectives to be fulfilled in this lab session. The first is to observe the effects of different disinfectants and antiseptics have on five different cultures using discs and plates. The second goal is to measure the sensitivity of a test microorganism to a variety of antibiotics using the Kirby-Bauer technique and a Mueller-Hinton agar plate. Procedure: Microbial Flora of the Hand 1. Record results in the data table in the lab manual. Microbes in the Food Supply 1. Examine TSA plates and using the dilution that nearly results in 30-300 colonies, calculate the number of organisms per gram of food sample using the following formula: a. original sample concentration = (# of colonies counted/volume plated) x (1/dilution counted). 2. Each pair needs to subculture a single colony of a specific type selected by the instructor into a TSA broth to be used for later. 3.
