Richard Duong MCRO 251 Lab 26 October 2010 Week 8 Pre-Lab Purpose : The objective of lab this week is to measure the variety of selective and differential medias, which inhibit the growth of certain types of bacteria while allowing the growth of others. Examples of which include Mannitol salt agar, Blood agar, and MacConkey agar. In this lab, we will test the following selective and differential medias: Triple Sugar Iron Agar, Citrate Utilization Medium, Motility-Indole-Ornithine Decarboxylase Medium, Urease Test Agar, and Oxidase Test. Procedure: 1. Complete observations from last lab in testing the effectiveness of disinfectants and antiseptics and antibiotic sensitivity testing by measuring the diameter zones of inhibition and recording them on the blackboard/table in lab manual. Compare and contrast. New Exercises 1. Label agar plates with drawer code, name, date, etc. and streak each plate with one of the organisms mentioned, sterilizing loops b/w quadrants: a. Mannitol salt agar: S. aureus, S. epidermidis, E. coli b. MacConkey agar: E. aerogenes, S. enterica, S. aureus c. Sheep blood agar: E. faecalis, S. viridians, S. aureus 2. Rubber band plates together and invert in incubation box. Triple Sugar Iron Agar (TSI)
