Richard Duong MCRO 251 Lab 10 November 2010 Week 10-11 Pre-Lab Purpose: This week’s experiment is designed to fulfill two purposes-to introduce Enzyme Linked Immunosorbent Assays (ELISAs), and to start the identification of our unknown cultures (one pure culture upper respiratory tract unknown and two unknown enteric organisms). Procedure: 1. Complete exercises from last week by observing the cultures as well as the additional demonstration plates of Streptococcus viridans, Streptococcus pneumoniae, Streptococcus pyogenes and Streptococcus agalactiae. 2. Observe Microbial Flora of the Throat plates for types of bacteria/hemolysis. ELISA 1. Label microtiter strip with appropriate information, cross out the underside of the six unused wells. a. Do not label wells 1-10 on bottom. 2. Use a single large Pasteur pipet to completely fill remaining 10 wells with 2% antigen solution. 3. Place in humidified tray where it will be kept at 4°C until two weeks later.
