Bioengineering 167L: Capstone Design I Lab 1: Introduction to a Bioengineering Laboratory 1.1 Pipetting and Weighing The pipette is a key tool in the cell biology laboratory for transporting a measured volume of liquid. In addition, laboratory practice should include an awareness of accuracy of measurements and knowledge of methods of assessing error and identifying sources of error. Although you will share your bench space and pipettes with a partner, you should each gather your own separate set of data. In this lab, you will be weighing tubes before and after pipetting a precise amount (mass) of water. Reagents 50 ml water Supplies Pipette tips for 1000 μ L, 100 μ L, and 10 μ L pipettes 2 ml microfuge tubes Equipment Pipettes, size: P1000, P100, P10 Balance 1.2 Procedure 1. Use latex gloves for this experiment in order to practice handling samples. 2. Obtain microtube rack and 9 empty 2 ml microtubes. 3. Label tubes with numbers 1 to 9 and your initials. 4. Obtain approximately 10 ml of water in a beaker. 5. Do not pipette or weigh until you’ve received instruction on the devices! Note: Pipettes are labeled by maximum (1000, 100, and 10 μ L). The minimum amount for each is ten percent of the maximum - pipetting amounts smaller than this will damage the pipette Liquid Loading Press the pipette down to the first stop (most of the way) Insert tip in liquid and slowly let the plunger slide back into the rest position Liquid Dispensing Transfer the pipette to the new container Press the pipette plunger down the first stop, then slightly harder until it reaches the second stop to unload
