Bioengineering 167L Lab 5 Process: Fluorescent microscopy of patterned surfaces. Start making new PDMS stamps Required Equipment: Fluorescent microscope Chemicals: Patterned surface (from Lab 4) Sylgard 184 base Sylgard 184 curing agent Supplies: Weigh boat, mixing utensil, acrylic mold SAFETY: Wear gloves, safety glasses, and lab coats. Do not gaze into the plasma cleaner for prolonged periods of time. PURPOSE: The objective of Labs 5 is image the adsorbed protein with the fluorescent microscope. Also we will make new PDMS devices (two) that will be used in contact angle measurements in lab 6. PROCEDURE : Fluorescence microscopy 1. Preparing the sample. Retrieve your sample from the refrigerator. 2. The fluorescent lamp will be left on because it may not be turned on until 30 minutes after it was turned off to prevent explosion or damage to the bulb. 3. The white light will be turned off and the software will be closed. Turn on the light switch located on the microscope. Reference the equipment guide for help at any time. 4.
