Bioengineering 167L Labs 9 & 10 Mammalian Cell Culture on 2-D Well Plates and in 3-D Encapsulation In this lab, you will learn how to seed cells in a well plate and perform a viability assay that can be used to quantitatively compare well-being of cells under different environments. You will also explore a method to culture cells in 3-dimensional environments that are more in vivo -like. Basic outline for lab 9 and 10 Laboratory 9 o TA s demo for Making alginate beads. o Per group: Encapsulate cells inside alginate beads. Each group will make its own alginate solution. TA will provide 0.1M CaCl 2 solution. Students will make three samples to encapsulate cells in alginate beads according to the bottom table. Sample 1 Sample 2 Sample 3 Volume of the cell suspension solution to add (ml) 3.0 2.0 1.0 How samples will be used in lab 10 Live/ Dead Stain Passage cells for next time (2 to 10 ratio or higher if necessary) o Cleaning up and dealing with the biohazard waste. For the solutions, aspirate or bleach until each solution reaches approximately 10 volume% bleach, wait for 10 minutes, and dump the solution down the drain with running water. Solid wastes will be taken by the TA to the autoclave site. Laboratory 10 o Per Group: Visualize the cell distribution within selected beads Apply the live/dead stain (TA will provide the dye mixture). View cells under the fluorescent microscope. Passage cells for next time (1 to 20 ratio or higher if necessary)
