The methodical instruction for lesson № 23 By biochemistry for students 2 course by specialty "General medicine " from September, 15 till September, 19th 2008 - 2009 year. The theme: Definition of normal and pathological components of urine. The main questions of the theme: 1 . Definition of physical and chemical properties of urine. 2. Definition in urine of proteins, blood, acetone, bilious pigments and glucose. 3. Definition of calcium and magnesium in the urine. 4. Definition of creatinin in the urine. Work N 1 Definition of bile pigments in urine (bilirubin). Course of work: Test on the bilirubin with solution of iodine. In tube pour in 3-4 ml of urine and carefully add 1% solution of iodine. At presence of bilirubin is formed the green ring between liquid. Definition of physic-chemical characteristics of urine. 1. Definition of color of urine. The normal urine has yellow color. The pathological components of urine can change the color. At presence of blood the color of urine will be red. At presence of bile pigments the color of urine will be yellow -green or yellow-brown. 2. The clarity . The urine of healthy human is clear. At the pathology the urine may be cloudy as a result of fall sediment (phosphate, and urate), and appearance in the urine of pus. N 3. Definition of specific gravity (urometer). Work 2 The find of ketone bodies in the urine. There are acetoacetate, ß - hydroxy butyrate and acetone. The ketone bodies are appeared in a urine by infringement of lipids and carbohydrates metabolism, diabetes and starvation (hunger). a/ The test by lugol on the acetone course of work: In tube pour in 2 ml of urine add 3-4 drops 10% solution of sodium nitroprusside and add 2-3 drops 10% solution NaOH. As a result appear the red-brown colour. If add 3-4 drops CH3 COOH the colour will be red. If acetone is absent in urine by addition CH3 COOH the red colour is changed to yellow. b/ The Lieben’s test on the acetone. Course of work: In tube pour in 2 ml urine add 3-4 drops 10% solution NaOH then add 4-5 drops solution of lugol till appearance of the slightly -yellow color. At presence of acetone the liquid becomes cloudy as a result of formation of slightly -yellow sediment of iodoformium. It has a characteristic smell.
