Competitive inhibition. The effect of a competitive inhibitor on an enzyme that obeys Michealis-Menten kinetics is to increase the observed K M , but not change the V max . In the Lineweaver-Burke plot of this data, the y-intercept corresponds to 1/V max and the x- intercept corresponds to -1/K M . For this type of plot, the y-intercept should be the same for all concentrations of a competitive inhibitor, because V max (and therefore 1/V max ) doesn’t change, which is what the plot above demonstrates. The x-intercept should be closer to zero with increasing concentrations of competitive inhibitor because as K M gets larger, -1/K M gets smaller in magnitude. This trend is also demonstrated in the plot above.
