BIOL430 Handout April 7 th , 2010 Mariam and Joanna SEPA-1 Mediates the Specific Recognition and Degradation of P Granule Components by Autophagy in C. elegans Key terms: Autophagy: is a catabolic process that delivers cytoplasmic material to the lysosomes for degradation. It regulates the degradation of long-lived cellular structures and proteins. It prevents the formation of protein aggregates which can be toxic to the animal. Autophagosome: is a double membrane vesicle that encloses cytoplasmic cargo to be delivered to the lysosomes for degradation. The formation of autophagosomes occurs during the process of autophagy. P-granules: A type of germ granules found in C. elegans . Cytoplasmic determinates in germ cells. PGL granules: contain multiple germ P granule components, such as PGL-1 and PGL-3. The Proteins: ATG18: Plays a role in the autophagy retrieval complex. LGG-1: Found on autophagosomes, binds to SEPA-1, and thus might play a role

Unformatted text preview: in recruiting SEPA-1 to the autophagosome. PGL-1 and PGL-3: Components of PGL granules. SEPA-1 (s uppressor of e ctopic P granule in a utophagy mutants): binds to LGG-1, required for PGL granule formation, found in somatic cells. Point form summary: BIOL430 Handout April 7 th , 2010 Mariam and Joanna 1) Autophagy degrades PGL granules in somatic cells. 2) PGL granules form in a gradual temporal fashion in somatic cells of autophagy mutants. 3) PGL granule components are maternally derived 4) sepa-1 is essential for PGL granule formation. 5) sepa-1 is essential for PGL component degradation by autophagy. 6) SEPA-1 is present in high levels during the 20-100 cell stage then decreases dramatically. 7) SEPA-1 is degraded by autophagy. 8) SEPA -1 and PGL components (PGL-1 and PGL-3) co-localize. 9) SEPA-1 and autophagy components (autophagosome) co-localize ....
