Introduction: The purpose of Research Paper is to use the Standardized Coliform Most Probable Number Test to exam the observable difference of the Most Probable Number of coliform bacteria in the water sample from the Elk River and the Kanawha River, then compare them with another river outside of the U.S. and finally get the results of coliform number rate to identify the cleanness level for each river. Coliform bacteria are a kind of parasitical bacteria in the human and animal large intestine. There are a great deal of coliform bacteria attached to human and animal excrements. Therefore, in the water purification and sewage disposal area, the coliform bacteria had been chosen for the index species in the water pollution level over a long period of time (Gleeson, 1997). The number of coliform bacteria number will measure how much human or animal excrement is in the water. The measurement standard used is known as the coliform bacteria index (Laskin, 1984). The reason one uses coliform bacteria as the standard test is because there is a larger number in human and animal’s excrement. In addition, coliform bacteria are harmless in most of situations and it will not harm people during the examination (Thomas and Simon, 2005). The best way to test the water quality of rivers, therefore, is the Standardized Coliform Most Probable Number Test. The experiment will use the hypothesis proposed which is that there is an observable difference in the most probable number of coliform bacteria in samples of water taken from the Elk River, the Kanawha River and the Nile River. In this paper, the test is based on the West Virginia Department of Environmental Protection standard. This is a standard use to measure maximum allowable level for fecal coliform in West Virginia. during the month. Then the river would be considered meets the standard. In opposite, if the coliform number is higher than 400/100ml, then that river will be considered not meets the standard. The two rivers in examination were all located in the State of West Virginia. The Kanawha River is a branch of the Ohio River. The total length of the Kanawha River is 97 miles. The source of the Kanawha River is located in Gauley Bridge in Fayette County, where is also the location that the Kanawha River merged by the New River and the Gauley River. It flows to northwest, through the cities of Charleston and St. Albans .
