So this eliminates sample 2. Then if you look at the size of the 2 pieces, you'll see they are pretty similar in size (450 and 550) and that they are relatively big. This tells you that the sample with just EcoR1 cut must be #3. b) 150 bp You know if there are at least three pieces there must have been more than just EcoR1 cuts. So if both enzymes cut the plasmid, you get 4 pieces. The sizes are 400, 400, 150, and 50. The 400s would be in the same place so they would make the same band, so the intermediate one would have to be the 150 nucleotide piece....
