Jlmarsh BioD137 Cancer ~1/3 of population dies of cancer in developed world Cancer is clonal When normal cells are polymorphic for any marker, tumor cells are monomorphic e.g. Abnormal karyotype shared by all the cancer cells CML t9;22 All tumor cells express same polymorphic version of G6PD x-inactivated gene All leukemia cells contain same kappa chain rearrangment Tumor cells are different than surrounding normal cells ways to get cancer Dominant ways: oncogenes 1- transforming retroviruses- misactivation of a piggy backed growth promoting gene by retrovirus = GOF by misexpression 2 – Activating point mutations. E.g. Ras mutations block conversion to inactive form. 3- activation by amplification. Often during chemo 4 – activating translocations/inversions/deletions etc. Chimeric proteins and inappropriate regulation. E.g. CML and t(9;22) Philadelphia chromosome BUT thought to need multiple mutations to get cancer so why are familial cancers so devoid of germline mutations in known proto-oncogenes? Perhaps order matters??
