Introduction The purpose of this experiment is to view enzymatic activity in PMA as well as DMSO treated HL-60 cells through a Zymography Test. MMP’s degrade the extracellular matrix which allows material such as macrophages to pass through. The extracellular matrix’s function is to holds cells and tissues in place. MMP-9 has multiple substrates but the one we will be focusing on is gelatin. By using gelatin as our substrate we should be able to see the function of MMP-9 in certain cell types. Our hypothesis is PMA cells should express a clearer band in the gel due to the presence of MMP-9 protein while DMSO cells will show a light or barely visible band due to the absence of the MMP-9 protein. Materials and Methods To begin this experiment we obtained 500 microliters of cell suspension in a micro centrifuge tube. Once the cells are obtained we centrifuged the cells for approximately fifteen seconds then discarded the supernatant. Next, we added .5ml of PBS to wash the cell pellet and the re-suspended the cell pellet. After re-suspension
