Necessary to create two cerebral hemispheres and optic lobes in anterior neural plate b. Disruption of Shh growth factor signaling leads to cyclopean embryo b.i. Sheep ate some sort of weed with jervine that blocks cholesterol synthesis b.ii. Teratology – study of birth defects B. Cholesterol a. Must be added to Shh for it to diffuse in a medial zone and block formation of: a.i. Pax6 : Phylogenetically highly conserved transcription factor involved in eye formation of organisms ranging from protostomes to deuterostomes (drosophila to vertebrates) C. Face duplications, including eyes, also possible D. Examples: a. Shh makes two Pax6 expression zones form in the diencephalon a.i. One central Pax6 expression area forms in Shh KO mouse leading to cyclopia b. Eyeless in flies formed by defective Pax6 gene – compound eye shown here doesn’t form
