Protein Sequencing The amino acid sequence is key to understanding the functions of a protein. Amino acid composition was initially determined using a complete acid hydrolysis of the peptide bonds Later, proteins were N-terminally sequenced using Edman degradation (see text). Proteins can now be sequenced and identified by mass spectrometry. Protein sequences today are most often deduced through DNA sequencing: This is much faster and easier.
