I THE NERVOUS SYSTEM This lab covers some basic anatomy of the nervous system, from the cellular level all the way up to large structural areas in the mammalian brain. Some things you will examine are in the Peripheral Nervous System (PNS) and some in the Central Nervous System (CNS). Some structures are named differently, depending on where they are located. For instance, in the CNS collections of neurons are called nuclei , but in the PNS, collections of neurons are called ganglia . Also, collections of axons are called tracts in the CNS, and nerves in the PNS. A. THE COMPOUND MICROSCOPE Microscopes are instruments used to produce magnified visual or photographic images of objects too small to be thoroughly examined by the eye alone. For many years, the biological and medical sciences have relied heavily on microscopy to solve problems relating to the overall morphological features of specimens as well as a quantitative tool for recording specific optical features and data. In this respect, the microscope has proven useful in countless investigations into the mysteries of life. The most widely used microscopes are optical microscopes, which use visible light to create a magnified image of an object. The compound microscope is one of the most common optical microscopes. It gets its name because it uses two lenses, an objective lens and an ocular lens mounted at opposite ends of a closed tube, which provides greater magnification than is possible with a single lens. A compound light microscope consists of a light source to illuminate the object to be observed, the magnifying lenses, a flat stage to hold the specimen, and some means for moving the specimen closer and further from the lenses, which changes the focus. The microscope is an expensive precision instrument that requires special care and handling. In this laboratory, you will learn the parts of the microscope, the functions of those parts, and the proper use and care of a microscope. Ordinarily, specimens are transparent and are mounted on slides. The stage has a small hole through which light can pass from a light source mounted underneath the stage. Objects or specimens are generally prepared in one of two ways. Prepared or permanent slides are made to last a long time. Temporary or wet mound slides are made to last only a short time, usually for a few hours at the most. 1. Care of the microscope Use both hands to lift and carry your microscope in a vertical position. Grasp the arm with one hand, and place the other hand under the base. Always place the microscope down very gently. Never remove any of the lenses. Only use lens paper provided by your instructor to clean the lenses. Do not use any chemical or water near the microscope.
