Bio 205L EXERCISE 3 Prep Sheet for LABORATORY PRACTICAL EXAM Procedures and Techniques learned during this laboratory: If the same technique is used in multiple sections of a lab, it does not need to be listed multiple times. ( For the lab practical exam, be sure you know how to properly perform each technique). 1. Section 3.1: Brightfield observations of human blood cells on permanent mount using oil immersion. a. Focus prepared slide under 100X magnification for light intensity, Kohler illumination, and ocular separation distance. b. Use stage travel knobs to find and identify the specimen. c. Follow proper procedure to bring a high dry objective into alignment and focus on specimen. d. Follow proper procedure to add oil to the slide and rotate in the oil immersion lens. e. With an oil immersion lens in place, use only the fine focus knob. f. Make a drawing of a white blood cell. g. Remove all immersion oil from the objective lens. 2. Section 3.2 : Brightfield observations of diatom frustules on permanent mounts a. Make low power observations of diatom frustules at 10x, then increase to 40x. b. Measure and record sizes of diatom frustules at 100x or 400x total magnification.
