Unformatted text preview: e bulb. Turn the light on. At one minute intervals, take the cuvette away from the lamp and measure the OD600 with the spectrometer. Always make sure the cuvette is aligned correctly. Read the absorbance value displayed in the box at the lower left. Manually record the absorption value in the worksheet below (it may fluctuate slightly, so count to five and write down the value then). 5. Repeat exposure to the light bulb light for 1 minute more and record OD600 again, and continue until it stops changing.
