Remember to keep these 5 sample tubes shielded from the light as you prepare them, and mix them well with a 1000µL. Record the initial OD600 absorption of all 5 cuvettes in the worksheet below. Always make sure cuvettes are aligned correctly in the spectrometer. These initial values should be very close if you have mixed them correctly. 2. Place the cuvettes at 20 cm intervals of distance from the lamp between 20 and 100 cm, and turn on the lamp. Align the tubes to that your labels face away from the lamp and the clear windows face toward the lamp. Make sure no cuvette is in the shadow of any cuvette that is closer to your lamp.
