Unformatted text preview: µl chloroplast suspension (15 mg chlorophyll) 2.9 ml phosphate buffer Mix until homogenous with a 1000µL pipetter. 2. Calibrate the Vernier Spectrophotometer at 600 nm. Click on the Configure Spectrometer button above the graph display. In the dialog box that appears, switch from Absorbance 21 Exercise 2 vs. Photosynthesis Wavelength mode to Absorbance vs. Time mode. You will then be asked if you want to save the data from the previous experiment. Since you have saved a PDF of your graph, you do not need to save the spectrum data. Set the wavelength to be measured to 600nm. First click on the Clear Selection button near the bottom of the dialog window, then scroll to 600nm and check the box next to it. Click OK.
