Bio Lab Study Guide Lab Techniques Pipettes o Micropipettes- small volume transfers Set volume on adjustment rings Use disposable tip Depress plunger to 1 st stop point then place in liquid and release o Volumetric- single volume precisely Amount indicated on top of pipet Draw up past engraved ring then release so meniscus is at ring o Mohr (measuring)- graduated but stop at baseline before narrowing Meniscus must be precisely on a calibration mark both at the beginning and end of the transfer o Serological- draw up fluid with meniscus precisely on correct mark Spectrophotometer o Turn it on o Wait 15 minutes o Set to appropriate wavelength o Set to transmittance and set at 0 o Clean outside of tube and put in o Set to 100 transmittance o Remove blank and put in first test tube (cleaning outside first) o Switch to absorbance Enzymes Determine rate of enzyme catalyzed reaction by examining the decrease of reactant or increase of product Hypothesis o Null- This will not affect this o Alternate- This will affect this Factors that impact rate of reaction………temp, structure of substrate, pH, concentration
