Two stages of synapse formation: 1. Recognition of postsynaptic sites and initial accumulation of synaptic vesicles. Synapses can be on dendrite, cell body, axon, muscle fiber… Adhesive factors : eg. Cadherin 2. Initiation of pre- and post synaptic specialization. Active zone : the site of synaptic vesicle docking and neurotransmitter release. Postsynaptic density : helps to localize neurotransmitter receptors/channels and signaling proteins to facilitate postsynaptic response to the neurotransmitter. Inductive factors : eg. neurexin/neuroligin

Neurexin-neuroligin interaction: -- important for synapse specialization. - - Neurexin helps to localize synaptic vesicles, docking/fusion proteins at active zone. - - Neuroligin promotes clustering of receptors and channels of postsynaptic density. Similar molecules used in axon growth are also used in synapse formation.

Specificity at synaptic sites: Different synaptic sites have different complements of adhesion molecules. eg1. Drosophila DSCAM (d own s yndrome c ell a dhesion m olecule) has ~38,000 alternative splicing products. An exon is any region of DNA within a gene that is transcribed to the final messenger RNA (mRNA).
