Lab 4: Apoptosis Zachary Bergman Partner: Michael Roche TA: Sujit Jangam Bio 210-3 Introduction The goal of this lab is to study the effects of apoptosis compared to necrosis in the death of a cell line called HCT116, derived from human colon carcinoma cells, which has a mutation in p53, a common cause of cancer. Apoptosis is the systematic killing and removal of faulty or old cells. Necrosis is the uncontrolled death of cells that is a chaotic process which is harmful to the organism and all neighboring cells. The p53 gene controls apoptosis by becoming phosphorylated and triggering the production of proteases called caspases. A mutation in this gene causes the cells to grow uncontrollably without dying creating tumors. This leads us to believe that the three p53 assays we will be running on the wildtype and mutant stands all will have more growth in the mutant strands as apoptosis will not occur. Results Please see attached data tables and agarose gel picture. Discussion 1. The protein topoisomerase creates nicks in the polypeptide backbone of DNA allowing the two strands to rotate around one another during synthesis. The addition of Doxorunicin inhibits this protein and greatly alters the efficiency of the cell. Without being able to unravel at certain parts of the DNA during replication, the entire strand of DNA would have to be unraveled to
