Sources- The exam is open book//open source.- You may not discuss it with anyone.- Please refer questions to Alejandro, or to any of the four lecturers. Pages- Start each question on a new page and put your name at the top of each page you submit.- You can write on the pages provided or use your own blank sheets.- Don’t exceed page limits; limits assume use of double line spacing.- Some questions request you show all your work – partial credit may be given.- Illegible answers will receive no credit. Point Distribution- The exam totals 100 points Questions 1-3 20 pts Questions 4-6 10 pts Questions 7-8 35 pts Questions 9-10 35 pts. Cell Biology (20 pts) 1. (1/2 page maximum) In the axon, anterograde transport is accomplished via kinesin family plus-end motors and retrograde transport is mediated by the dynein minus-end direct motor. Things are more complicated in dendrites due to differences in the cytoskeleton. a. What is different (2 pts)? b. What problem(s) does this cause (2 pts)? c. Provide a plausible explanation for why cargos are still able to reach distal dendrites (2 pts). Extra credit for providing more than one plausible explanation. 2. (1/3 page maximum) Describe the major differences between the cell biological life-cycle of a cholinergic synaptic vesicle and a Neuropepdtide-Y dense core vesicle (4 pts). 3. (1/2 page maximum) `Your have just used your new antibodies against RCP-1 (Really Cool Protein) and use it to perform immunohistochemistry on cultured hippocamppal neurons at 1, 5, 12, and 15 days in culture. You see very little staining at 1, 5, and 12 days in culture and then robust staining that co-localizes with approximately 1/4th of glutamate receptor puncta at 15 days in culture. a. Propose a hypothesis to account for the development of this delayed and selective localization pattern (4pts) b. Outline three simple experiments you might do to test the hypothesis (3 x 2 pts). 4. (1/3 page maximum): Describe three functions of astrocytes in the CNS. Define any specializations (morphological/ molecular) that supports these functions. (3 pts) 5. (1/3 page maximum): What mechanisms have evolved to increase the rate of action potential propagation along axons? Discuss why the vertebrate lineage might have favored one mechanism over another. (3 pts) 6. (1/3 page maximum): Discuss similarities (at least 2) and differences (at least 2) between oligodendrocytes and Schwann cells.
