Pre-lab 4 - Bacterial Isolation and Titration of Bacteriophage February 1-3 (To be turned in PRIOR to start of recitation for lab 4) Name: __TD Khang_________________________________________ Objective: Perform another streak plate of the bacteria prepared last week. Use the following  equation, Original concentration of phage = (# of PFU/volume plated) x (1/dilution of the tube  counted), to determine how much bacteria can grow from a phage infecting a host cell. Procedure: 1. Perform the streaking procedure from Lab 3. 2. Label all pipets, broth tubes, plates and soft agar. Make 10^-1, 10^-2, and 10^-3 dilutions  of the phage. 3. Flick the tube of phage to remix. 4. Keep the pipettes sterile and use them only for correct purposes. 5. Get the 3 broth tubes of diluted phages. Get 1.0 ml of phage from the stock solution with a  pipette and put it in the 10^-1 tube. Mix the tube.
