Lab Session 10: ID of Gram + Cocci, Throat Flora March 22 - 24 (To be turned in PRIOR to start of recitation for lab 10) Name: _______Td Khang_______________________________ Objective: Analyze and record results of the selective and differential media. Observe and record the results for the Triple Sugar Iron Agar. Observe and record the results for the citrate utilization medium. Observe the MIO Medium for motility and ODC and then test for the indole. Perform a catalase test to test for the production of catalase in a bacteria. Perform a coagulase test to identify staphylococcus. Prepare four test for catalase negative streptococci. Observe and record results of microorganism located in the throat. Procedure for MIO medium: 1. Observe the plate for motility. 2. Observe the plate for ornithine decarboxylase. 3. Test for indole by putting drops of Kovacs’ reagent. Procedure for catalase test: 1. Get a loop full of bacteria.

Procedure for Coagulase test 1. Put a drop of plasma on a slide then mix with a loop of Staph. Aureus. 2. Mix with inoculating loop and repeat procedure with Staph epidermidis Procedure for identification of staphylococcus: 1. Label plate with correct info and streak with Enterococcus faecalis 2. Label plate with correct info and streak with S. pyogenes 3. Sterilize forceps and add an A disc to second quadrant of S. pyrogenes streak plate. 4. Label plate with correct info and streak with S. pnuemoniae 5. Sterilize forceps and add a P disc to second quadrant of S. pnuemoniae streak plate. 6. Streak a slant with s. pyrogenes 7. Streak remaining slants with E. Faecalis 8. Put plates in correct location for incubation...
