30 1. Diisi masing-masing 100 mL media ke dalam 2 erlenmeyer 250 mL. 2. Diinokulasikan ke dalam masing-masing erlenmeyer satu biakan murni dari agar miring 24 jam. 3. Dimasukkan satu erlenmeyer ke dalam shaker untuk dilakukan pengocokan pada suhu kamar. Dicatat suhunya. Sedangkan erlenmeyer yang lain tanpa dilakukan pengocokan pada suhu kamar.
