Emily Lin 3/1/11 Topic 6: DNA Replication F 14.2-14.4 Topic 6 discusses how researchers solved mysteries of the structure of DNA and how it’s repaired. DNA is a long, linear polymer made up of monomers called deoxyribonucleotides, which consist of a deoxyribose molecule, a phosphate group and a nitrogenous base. These deoxyribonucleotides link together into a polymer when a phosphodiester bond forms between the hydroxyl group on the 3’m carbon and the phosphate group attached to the 5’ end. The primary structure of a DNA molecule has a backbone made of the sugar and phosphate groups of deoxyribonucleotides and a serious of nitrogen containing bases that project from the back bone. DNA has a hydroxyl group on the 3’ end and a phosphate on the 5’ end, so it is polar. The structure is overall an anti-parallel double helix, which is stabilized by the hydrogen bonds made between the Adenine, Thymine and Cytosine, Guanine according to complementary base pairing rules. Watson and Crick suggested that the existing strands of DNA serve as a template for the production of new strands, with bases added to the new strands according to complementary base pairing. The Meselson-Stahl Experiment used E.coli and tagged parental and daughter strands of DNA that would make them distinguishable from each other to see if replication was conservative, semi conservative or dispersive. They grew the cells in one of two isotopes of nitrogen, ones with 14 and 15 neutrons, so that during centrifugation, strands would behave differently. If conservative, double stranded DNA with
